Specific Topics:

The primary focus of our research group is the study of mechanistically interesting enzyme-catalyzed reactions including alkane formation, S.A.M. – derived C-methylation of unactivated double bonds (cyclopropane synthase), hydroxylation (Alk B w-hydroxylase) and dehydrogenation of unactivated hydrocarbon chains (fatty acid desaturases and acetylenases).  We have developed versatile active-site probes which allow one to elucidate the stereochemical and cryptoregiochemical details of this important set of biological reactions. This approach has resulted in our participation in a number of local, national and international collaborations.  Spinoff projects include stereochemical analysis of sulfoxides  and applications of 1H-decoupled 19F NMR.  Reviews of work in the desaturase area have been commissioned by the editors of Current Opinion in Chemical Biology, Phytochemical Reviews, Prostaglandins, Leukotrienes & Essential Fatty Acids, Natural Products Reports , Tetrahedron Asymmetry, Wiley Encyclopedia of Chemical Biology and Comprehensive Natural Products Chemistry.  Our results have relevance to several areas related to lipid metabolism such as the metabolic syndrome, bioremediation, plant defence strategies, insect communication and biofuels.
