Angiogenesis driven extracellular matrix remodeling of 3D bioprinted vascular networks<\/span><\/strong><\/em><\/p>\nAngiogenesis\u00a0plays a\u00a0pivotal role\u00a0in development and tissue growth, as well as in pathological conditions such as cancer. Being able to understand the basic mechanisms involved in the\u00a0vascularization\u00a0of tissues and angiogenic network formation provides a window to advance the development of\u00a0in vitro\u00a0tissue models and enhance tissue engineering applications. In this study, we leveraged a novel microfluidic-based three dimensional (3D) bioprinting technology and alginate-collagen type I (AGC) bioink, to develop a\u00a03D bioprinting\u00a0strategy to enable the\u00a0biofabrication\u00a0of complex angiogenic networks within the 3D structure. These networks were comprised of\u00a0simian vacuolating virus\u00a040 (SV40) transformed adult rat brain\u00a0endothelial cell\u00a0(SV-ARBEC)-laden hydrogel rings. With mechanical properties relevant for\u00a0vascular tissue\u00a0engineering applications, these bioprinted constructs formed spontaneous\u00a0vascular networks, reminiscent of\u00a0anisotropic\u00a0tissue-like structures, while retaining high\u00a0cellular viability. The vascular network formation was accompanied by\u00a0extracellular matrix\u00a0(ECM) remodeling, confirming sequential SV-ARBEC mediated\u00a0collagen type\u00a0I fiber deposition and reorganization. Treatment with broad spectrum matrix metalloproteinase (MMP) inhibitor supressed SV-ARBEC\u00a0angiogenic sprouting, highlighting requirements of ECM remodeling in angiogenic network formation.
